Brain insulin resistance contributes to cognitive decline in Alzheimer's disease

Insulin resistance in the brain precedes and contributes to cognitive decline above and beyond other known causes of Alzheimer's disease, according to a new study by researchers from the Perelman School of Medicine at the University of Pennsylvania. Insulin is an important hormone in many bodily functions, including the health of brain cells. The team identified extensive abnormalities in the activity of two major signaling pathways for insulin and insulin-like growth factor in non-diabetic people with Alzheimer's disease. These pathways could be targeted with new or existing medicines to potentially help resensitize the brain to insulin and possibly slow down or even improve cognitive decline.

This is the first study to directly demonstrate that insulin resistance occurs in the brains of people with Alzheimer's disease. The study is now online in the Journal of Clinical Investigation.

"Our research clearly shows that the brain's ability to respond to insulin, which is important for normal brain function, is going offline at some point. Insulin in the brain not only modulates glucose uptake, but also promotes the health of brain cells ? their growth, survival, remodeling, and normal functioning. We believe that brain insulin resistance may be an important contributor to the cognitive decline associated with Alzheimer's disease," said senior author, Steven E. Arnold, MD, professor of Psychiatry and Neurology. Arnold is also the director of the Penn Memory Center, a National Institute on Aging-designated Alzheimer's Disease Core Center. "If we can prevent brain insulin resistance from occurring, or re-sensitize brain cells to insulin with any of the currently available insulin-sensitizing diabetes medicines, we may be able to slow down, prevent, or perhaps even improve cognitive decline.

The risk of developing Alzheimer's disease is increased by 50 percent in people with diabetes. Type 2 diabetes is due to insulin resistance and accounts for 90 percent of all diabetes. The defining clinical feature of Type 2 diabetes (and Type 1 "juvenile" diabetes) is hyperglycemia - high levels of sugar in the blood ? but there is no evidence that the brain in Alzheimer's is hyperglycemic. Insulin acts differently in the brain than in the rest of the body. Researchers found that insulin resistance of the brain occurs in Alzheimer's disease independent of whether someone has diabetes, by excluding people with a history of diabetes from this study.

The investigators used samples of postmortem brain tissue from non-diabetics who had died with Alzheimer's disease, stimulated the tissue with insulin, and measured how much the insulin activated various proteins in the insulin-signaling pathways. There was less insulin activation in Alzheimer's cases than in tissue from people who had died without brain disease. Other proteins linked to insulin action in the brain were abnormal in Alzheimer's disease samples. These abnormalities were highly correlated with episodic memory and other cognitive disabilities in the Alzheimer's disease patients.

In tissue from people with Alzheimer's disease and mild cognitive impairment (MCI), researchers found that changes to a protein called insulin receptor substrate-1 (IRS-1 pS636/639 and pS616) in brain cells were linked to the severity of memory impairments regardless of age, sex, diabetes history, or apolipoprotein E (APOE) gene status. Levels of IRS-1 were also significantly associated with, but not likely to affect, the presence of amyloid beta plaques and neurofibrillary tangles, the signature markers of Alzheimer's disease. This suggests that insulin resistance contributes to cognitive decline independent of the classical pathology of Alzheimer's disease.

Researchers noted that three insulin-sensitizing medicines are already approved by the FDA for treatment of diabetes. These drugs readily cross the blood-brain barrier and may have therapeutic potential to correct insulin resistance in Alzheimer's disease and MCI. "Clinical trials would need to be conducted to determine the impact the drugs have on Alzheimer's disease and MCI in non-diabetic patients," said Dr. Arnold.

'Tuberculosis and TB/HIV/AIDS: Unanswered Questions and Controversies' launches at World TB Day Conference 'Tuberculosis -- Local and Global'

To coincide with World TB day (24th March), The Journal of Infectious Diseases (JID) publishes a special supplement: 'TUBERCULOSIS and TB/HIV/AIDS: Unanswered Questions and Controversies', guest edited by Professor Alimuddin 'Ali' Zumla (Division of Infection and Immunity, UCL) and Dr. Marco Schito (Henry M. Jackson Foundation for the Advancement of Military Medicine*).

The articles from the JID supplement will be presented, along with local tuberculosis research performed at UCL and the Royal Free Hospital, at a World TB Day Conference 'Tuberculosis: Local and Global'. The event will be hosted by UCL at the Royal Free Hospital in London on Friday March 23rd.

Worldwide, tuberculosis (TB) remains one of the most important causes of death from an infectious disease. TB and TB/HIV co-disease caused an estimated 1.4 million deaths in 2010 and there were 8.8 million incident cases of TB, with the highest rates occurring in sub-Saharan Africa and the largest number of cases in Asia. Sub-Saharan Africa continues to be disproportionately affected by the dual epidemics of TB and HIV. Moreover, the number-one cause of death for HIV-infected patients in sub-Saharan Africa is TB.

While recent TB control efforts have been substantial and progress in the development of new diagnostics, drugs and vaccines has accelerated, the implementation of coordinated TB/HIV treatment and prevention activities remains challenging. Despite nearly 20 years of World Health Organisation (WHO)-directed and coordinated activity, and 12 years of multidrug-resistant (MDR) tuberculosisspecific activity, "the response to the drug-resistant tuberculosis epidemic seems to be ineffectual, with projected rapid increase in the global incidence of Multi-drug resistant tuberculosis (MDR-TB)," said Professor Zumla. "It (MDR TB) continues to spread and now poses a major threat to TB control - recent reports of highly drug resistant TB from Iran and Bombay are worrisome," he continued.

According to the supplement's accompanying editorial, the main obstacle hindering progress toward achieving TB control seems to be the lack of creative and lateral thinking with unity of purpose. Historically, some widely held assumptions, dogma, and orthodoxy have prevailed, leading to polarization of thought, varying opinions, and continued debate within the scientific, medical, funding, and various other stakeholder communities. The need for open debate and fostering of innovation is reflected by the needs surrounding all aspects of tuberculosis and HIV control strategies.

This need has never been greater, especially in the current adverse global economic climate.

The JID supplement arises from the urgent need to focus attention on core problems and secure a broad spectrum of opinion from TB and HIV/AIDS experts from various geographical backgrounds. It comprises 23 articles written by 164 authors from all over the world.

This special volume critically highlights current controversies and unanswered questions in TB and TB/HIV issues affecting adults and children, and provides insights into newer and enhanced approaches to achieving the Millennium Development Goals for TB control. Specifically, the volume covers operational issues, challenges, and priority needs relating to a broad range of diagnostic, management, prevention, surveillance and related research issues. The array of priorities spans development, evaluation, and implementation of new drugs, diagnostics, and vaccines.

"Political commitment, regulatory cohesiveness, donor investments, and increased coordination among all stakeholders is necessary to make progress," says Professor Ali Zumla. "This includes upgrading of laboratory services for early and accurate diagnosis, development and validation of better diagnostic testing algorithms, effective treatment at points of care, and improved surveillance.

"This supplement also underlines preventable factors, such as crowded living conditions and under-nutrition that continue to make communities vulnerable to disease," continues Professor Zumla. "The influence of non-communicable diseases, such as smoking-related lung disease, diet-related diabetes mellitus, malnutrition, as well as alcohol and drug abuse, highlight the need for a multi-disciplinary approach in tackling both TB and HIV."

Despite increased funding for TB, several articles point to a serious shortfall which continues to persist. The financial difficulties faced by the Global Fund place recent gains in TB control and the numbers of HIV-infected persons on life-saving treatment at serious risk in many donor-reliant countries. The global fight against TB and HIV requires perseverance, assertiveness, and innovation, and clarity of intent well beyond current efforts. All authors in the supplement call on governmental, civil society, non-governmental groups, donors, and funders to work collectively to assist in global control of TB and, ultimately, its elimination.

(AP) ? Soldiers stormed the presidential palace of one of the few established democracies in this corner of Africa and announced Thursday that they had seized control of the country, ousting Mali's president just one month before he was due to step down at the end of his legal term.

Gunfire could be heard ringing throughout the capital, and the mutinous soldiers imposed a nationwide curfew, warning people to stay off the streets until further notice. A flight headed to the capital of Bamako was forced to make a U-turn in the air after the airport closed.

The soldiers said they are overthrowing the government because of its mishandling of an ethnic Tuareg insurgency in the country's north that began in mid-January. The rebellion grew when fighters who had supported the late Libyan leader Moammar Gadhafi returned home heavily armed. The unrest has forced tens of thousands to flee, and has prompted fierce criticism of Mali's government.

Early Thursday, a group of around 20 soldiers in military fatigues were shown on national television crowding around a desk, facing the camera.

They announced that Mali is now under the control of the military's National Committee for the Reestablishment of Democracy and the Restoration of the State, or CNRDR. They said they were suspending Mali's constitution and dissolving its institutions.

The soldiers complained that the civilian government had not done enough to combat a rebellion by the ethnic Tuareg separatists, who want to carve out a homeland in the country's northern desert.

"The CNRDR representing all the elements of the armed forces, defensive forces and security forces has decided to assume its responsibilities and end the incompetent and disavowed regime of (President) Amadou Toumani Toure," they said, reading from a statement. "The objective of the CNRDR does not in any way aim to confiscate power, and we solemnly swear to return power to a democratically elected president as soon as national unity and territorial integrity are established."

The West African regional bloc ECOWAS said it "strongly condemns the misguided actions of the mutineers." The group said it was "all the more reprehensible, coming amidst the ongoing regional and international efforts to seek a peaceful solution to the rebellion in the north of the country."

The soldiers' declaration also came just hours after the presidential palace had rushed to deny that a coup was in progress, issuing a Tweet that said: "There is no coup in Mali. There's just a mutiny."

The latest developments mark a major setback for one of the region's few established democracies. The ousted president came to power himself in a 1991 coup, but was hailed for handing power to civilians. A decade later, he won the 2002 democratic election. The 63-year-old Toure was due to step down next month after two, five-year terms.

His whereabouts were unknown on Thursday. Contacted by telephone, a soldier at the palace said that the president's bodyguards had failed to fight the renegade soldiers, who burst in. They searched the grounds looking for him, but could not find Toure.

Young soldiers on motorcycles were shooting in the air Thursday morning in the capital of Bamako. Businesses remained shuttered and there was little traffic as most stayed home following the coup announcement.

On Wednesday, Defense Minister Gen. Sadio Gassama had come to a military camp in the capital for an official visit but failed to address the grievances of the rank-and-file soldiers in his speech to the troops. The rebellion has claimed the lives of numerous soldiers, and those sent to fight say they are not given sufficient supplies, including arms or food.

Recruits started firing into the air Wednesday morning and by afternoon, troops had surrounded the state television station, yanking both the television and radio signals off the air for the rest of the day. By Wednesday evening, troops had started rioting at a military garrison located in the northern town of Gao, some 2,000 miles (3,200 kilometers) away.

A freelance journalist from Sweden who was driving to her hotel near the TV station in Bamako around 4 p.m. Wednesday said that trucks full of soldiers had surrounded the state broadcaster, where they set up machine guns facing the building.

"They came and started setting up checkpoints. There were military in the streets, stopping people," said Katarina Hoije. "When we reached our hotel which is just in front of the TV station, there were lots of military outside, and more cars kept arriving ? pickup trucks with soldiers on them."

The Tuaregs, a traditionally nomadic people spread across the Sahara Desert, have risen up against the central government in Mali several times since the country's independence from France in 1960.

The newest rebellion launched in mid-January broke years of relative peace, and has been fueled by the return of Tuaregs from Libya who had fought in Gadhafi's army. Refugees have spilled over into four of the countries neighboring Mali due to the uprising.

Thousands of Tuaregs moved from Mali to Libya over the decades beginning in the 1970s, and many joined special divisions of Gadhafi's military where they earned higher salaries than in Mali. A relationship developed between the Tuareg and Gadhafi, who claimed they had distant blood links.

When the Gadhafi regime fell last year, Tuareg troops smuggled Gadhafi family members to neighboring Algeria and Niger. After Gadhafi was killed in his hometown of Sirte in October, many Tuaregs no longer felt safe in Libya and began returning to Mali.

The government has not disclosed how many government soldiers have been killed. The toll is believed to be significant and in February, military widows led a protest, publicly grilling Toure on television over his handling of the rebellion.

Cats have been found weeks after they go missing. Since the conception of the micro-chip, dogs have been returned home long after their replacement has arrived.
When a pet goes missing it is vital to act fast and to never stop the search. Grab a powerful flashlight even during daylight hours, we learned this from watching ?CSI?. Search dark spaces. A frightened or injured cat will hide in dark spaces and will not come to you when called. An injured dog may also.

Immediately search your property thoroughly. Small dogs and cats can get into some incredibly strange places. Look in EVERY nook and cranny. Don?t assume that your pet would never crawl into a tiny space. Especially cats.. felines are a curious group. Look behind, under, and inside washing machines, clothes dryers, stoves, refrigerators, dishwashers, bookcases, behind water heaters, under furniture, under beds, in closets, in cabinets, in drain pipes, in sewer drains, in culvert pipes, under vehicles, under the hood of your car, in crawl spaces under the house, inside sheds and barns, etc. In the case of cats, also look in attic crawl spaces, on the roof, in roof gutters, and up in the trees.

Make some noise while you walk around the neighborhood! Animals can hear you from great distances. If your pet has a favorite ?squeaky toy? bring it along and use it to help you make familiar noises. It?s also important to stop regularly, be quiet, and listen for your pet to make a noise in reply. Especially at dawn and dusk.

Place strong-scented articles outside your home to attract your pet. Leaving the pets bed, a piece of your clothes or personal blanket. They will help to give the pet an anchor to stay near or on if he or she is lost when you are away from home or if you have to leave the last area where you saw your pet.

Put flyers everywhere. It is extremely important to post MANY flyers within at least a 1-mile radius of where it was lost. DO NOT PUT YOUR NAME OR ADDRESS ON YOUR FLYER! List the date and place your pet was lost, breed of dog or cat, sex, age, weight, color, markings, and your telephone number. Offer a reward, but don?t state the amount. It is very important to always withhold several identifying marks and characteristics of your lost pet. Beware of the scammer. Post the flyers at waist level on telephone poles and at eye level in such places as veterinary offices, pet shops, beauty shops, grocery stores, community bulletin boards, churches, pizza parlors, convenience stores, near schools, and on school bulletin boards, on windshields, in mailboxes, fire stations, Starbucks, food trucks and construction worksites. Examine your posted flyers frequently and replace the ones that are missing or damaged.

Walk the neighborhood and talk to everybody. Go to each house in the area where your pet was lost and talk to the residents; give them your flyer. If no one is home, leave the flyer attached to his or her door, don?t forget to bring tape. Talk to everybody you run across ie, neighborhood children and parents waiting at the school bus stop.
This also includes the paperboys, school crossing guards, neighborhood crime watch groups, garbage pick-up crews, postal workers, sanitation workers, construction workers, etc. Give them your flyer.
Place an ad in your local newspaper. Some will do this for free. Be sure to advertise in the Sunday edition as well as during the week. Also place an ad in any ?Penny Saver? type of publications. Check the newspaper ?found? ads every day. Also check online ?Lost and Found?. Most newspapers provide free ads to people who have found lost pets.

Call local veterinarian offices during the day. After 5 PM, call veterinarian emergency clinics. If an office has taken in or treated any animal that even remotely resembles your pet, VISIT THE OFFICE IN PERSON. Your description of your pet and their description of the same pet rarely match. YOU MUST GO SEE FOR YOURSELF!

Get phone numbers of local rescue organizations. The free Pet Press (at the library and pets stores) has current lists. Call each of the rescue organizations and ask for their help and find out if they have your pet. These groups generally network with each other and will pass the word about your case. VISIT your local Animal Control, humane societies, and animal shelters, including the ones in surrounding areas. You must actually visit the animal control and humane shelters every day or two. It works well if several family members can take turns visiting the shelters. Be sure to check all areas of the shelter, including the infirmary. Also be aware that dogs may be housed in the cat section and vice-versa.

Leave a picture of your pet and your phone number at each shelter, befriend them. Find out the holding period of each animal control and humane shelter. Be aware of how much time you have to claim your pet before it is euthanized!

Check shelters out of your area in person and online. A Good Samaritan may have taken your pet in only to have it escape a week or more later.

Don?t ever give up! Pets have been known to find their way back home after being lost for several months.

PREVENTION: Get your pet Micro-chipped, keep tags current.

Keep pet tags current and check that they are still in place and legible. A good idea is to write your phone number directly onto the collar. I do not remove my dog?s collar even when she is being bathed. I have heard the excuse that the lost pet was not wearing a collar and id because they were just bathed.

Your tag should have more then one phone number, your home number, cell, your vet etc.. I keep two collars and tags on my dogs as well as a microchip.

Get an ?Acme Dog Whistle?. Blow the whistle whenever you feed or give treats to your pet. Leave the whistle next to the treats so that you will remember to use it, this really works. The high-pitched sound from these whistles can carry up to a mile or more. Cats are attracted to this sound as well as dogs. (It is also a great way to teach a pet the ?come command?.)

Keep fence gates closed securely. Self closing gates area good idea. Always transport a cat in a carrier. Never take your cat to the Vet or anywhere else unless it is secured. A hand carried cat can bolt and hide if frightened by loud noises. When a cat is frightened in strange surroundings, especially with traffic noise around, it will hide and will not come to you.

If you have a dog who is an escape artist, try leaving a piece of broken leash hanging front their collar so that it will appear that the dog broken lose as opposed to being out for a walk.

NEVER, NEVER leave a choke collar on a dog when it is not attached to a leash. It will eventually become a HANGMAN?S NOOSE!

Have current photos of your pet.

A pillowcase is a quick way to transport cats or injured small animal.
As a final point, spay or neuter your pets! Both males and females will be much less likely to wander if ?fixed.?

Bangladesh's "teenage" brothels hold dark steroid secret

TANGAIL, Bangladesh (Reuters) - Their faces painted heavy with make-up, teenage girls in short, tight blouses and long petticoats loiter in squalid alleys, laughing and gesturing to potential clients who roam Tangail town's infamous red light area in the early evening.

There is no shortage of men looking for "company" in Kandapara slum, a labyrinth of tiny lanes - lined cheek-by-jowl with corrugated iron shacks - a few hours drive northeast of Bangladesh's capital, Dhaka.

But with rates as low as 50 taka (60 U.S. cents), the need to attract as many customers as possible is desperate - prompting a rising, yet dangerous, trend of steroid abuse among adolescent sex workers to "enhance" their appearance.

"There is a huge difference between my appearance now and the malnourished look of my childhood," says Hashi, 17, who was lured into the sex trade by a trafficker when she was 10 and sold to Kandapara's brothel, where she began taking steroids.

"I am healthier than before and fit to serve a lot of customers in a day. Sometimes up to 15," she says, placing a large black bindi, or dot used by Hindu women, between her perfectly shaped eyebrows.

She sits in her tiny room with a bed, a cooking stove and posters of Bollywood stars taped across the wall.

Hashi is one of around 900 sex workers - some as young as 12 - living a painful life of exploitation in Kandapara, not only bonded by debt and fear of stigma, but compelled to take the steroid, Oradexon, which brings more income but leaves dangerous side effects.

COW FATTENING DRUG

Also known as Dexamethasone, Oradexon treats inflammation and allergies in humans and is used by farmers to fatten livestock.

Charities say the over-the-counter drug is taken by 90 percent of sex workers in Kandapara and the other 14 legalized brothels across this impoverished South Asian nation.

The girls are first forced to take it by their madams, or "sardarnis", who run the brothels.

It increases their appetite, making them gain weight rapidly and giving the appearance that these poorly nourished teens are in fact healthy and older - attracting clients who prefer girls with "curves".

It also helps sardarnis keep the police away. The legal age for sex work in Bangladesh is 18.

The girls then continue to consume it, saying that it keeps them "strong and healthy", which in turn will help them get more clients in a day so they can earn enough to survive.

"My sardarni forced me to take a tablet. She beat me up and stopped giving food. She threatened me and reminded me about my loans," says Hashi, who has a four-year-old son staying with relatives, whom she has not seen for two years.

"In this brothel, customers always look for healthy girls. I take Oradexon. I need customers so I can pay my bills and loans. If I don't get any customers one day, I cannot eat in the next day. I wish to save some money for my son."

The story is the same with most of Kandapara's teenage sex workers, or "chukris".

Sold for as little as 20,000 taka ($245) by their poor, rural families to traffickers, they are then traded on to brothel sardarnis, who are former prostitutes themselves and keep the teenagers in bonded sex work.

The girls speak of being with up to 15 men in one day, but say their earnings are pocketed by their sardarnis, who tell them they have to work to pay off the money paid for them.

Many girls have been in Kandapara's brothel for years, yet due to their illiteracy, they have no idea whether their debts have been cleared and what their rights are.

Others, who have been left by their sardarnis because they are too old or not-profitable, are in principle free to leave but choose not to, fearful of the social exclusion they will face in the conservative, Muslim society outside of Kandapara.

BATTLE WITH BROTHELS

Oradexon, they say, keeps them going, even though there are known risks associated with its long-term use.

The steroid can cause diabetes, high blood pressure, skin rashes and headaches and is highly addictive, according to social activists.

It also weakens the immune system and leaves patients more susceptible to illnesses. There have been reports of young sex workers dying from over-use of the drug.

The small white pill is easily available in Kandapara's slums. It is sold without prescription for 15 taka (18 U.S. cents) for a strip of 10 at the tea and cigarette stalls blaring Bangladeshi pop music that populate a maze of open-sewer lanes.

"Steroids are life-saving as well as life-destroying drugs which are used by sex workers in poor countries," said Shipra Gowshami, a lawyer and human rights activist who works with sex workers in brothels in the central Bangladeshi town of Faridpur.

"A lack of awareness, easy availability and malpractices of quacks are some of the prime causes of why these drugs are being abused," Gowshami said.

In 2010, ActionAid Bangladesh began a campaign to promote awareness of the drug among sex workers. But they say they are facing a long fight in persuading not only the brothels to stop using it but also authorities to regulate it.

"There have been attempts to raise awareness on the negative impact of the use of such medicine but brothel owners, madams and pimps are a long way from withdrawing such practices," said Farah Kabir, country director for ActionAid Bangladesh.

"We have an uphill battle, yet it can be won. There needs to be greater regulation in the sale of such drugs. Government and the state must play an active role."

Obama marks St. Patrick's Day with pint in a pub

WASHINGTON (AP) ? His jacket was only moss green but his pint was true Guinness.

President Barack Obama tilted back a glass of the dark Irish brew Saturday, observing St. Patrick's Day at a boisterous Irish pub with his ancestral cousin from Moneygall, Ireland, at his side.

At the White House, the main South Lawn Fountain burbled green water. Nearby, workers prepared for a visit Tuesday by the Irish Prime Minister Enda Kenny.

The first family was putting on its Irish, a blood line that runs through Obama's veins.

Obama took his motorcade to the Dubliner Restaurant and Pub on a dazzling Saturday afternoon. He wore no Kelly green but his jacket was pierced with a button that read, "VIP GUEST ? Tell 'em Danny sent you." The president waded into a crowd ? some in leprechaun hats and others in dyed green hair ? at the entrance of the tavern near Washington's landmark Union Station.

He wished one reveler, Adam Joseph, a happy 29th birthday.

Reporters were ushered into the pub briefly, long enough to catch the president taking two sips of his beer. The thick foam stuck to his upper lip in a thin mustache. And he held Dubliner owner Danny Coleman's 21-month-old grandson, Danny, in his arms.

"Hey, Danny, who's this?" Obama asked, holding out his VIP GUEST button, which bore the child's image.

One of Obama's great-great-great grandfathers on his Kansas mother's side was Falmouth Kearney, a shoemaker who emigrated from Ireland to the United States in 1850. Last year, Obama visited his ancestral home of Moneygall, a small hamlet in Ireland, and was a hit when he drank a Guinness at the local pub.

On Saturday, the owner of that pub, Ollie Hayes, and Henry Healy, an eighth cousin to Obama and the closest relative still living in Moneygall, joined him barside at the Dubliner as his guests.

Some in the bar crowd chanted "Four more years!"

Coleman said the president finished his beer, and predicted that, at least on this day, Obama could count on the support of the millions of Americans who claim Irish ancestry.

On Tuesday, Obama and Vice President Biden will meet Kenny and attend a St. Patrick's Day lunch at the Capitol. Then the president and first lady Michelle Obama will host an evening reception at the White House.

As Obama left the Dubliner amid more shouts and calls of "Go Bulls!", the president offered lively patrons a bit of advice often unheeded on St. Patrick's Day.

"I expect you guys," he said, "to behave yourselves."

Meteorologist swears, walks off live TV

Technology is supposed to help us out at work, but when it gets in the way instead, it's easy to get frustrated. That happened Wednesday?to Rachael Plath, meteorologist at KRDO-TV in Denver.

In the video clip below, Plath is seen giving the weather forecast on air, when the graphics behind her change, apparently prematurely.?Plath suddenly exclaims "Son of a b----, why is this happening!" and walks off the set.

The video was quickly picked up by The Huffington Post and the U.K.'s Daily Mail, leading to numerous viewers posting sympathetic notes on Plath's Facebook page.?

Wrote Robert Borges, "No need to ever feel bad or apologize for being a human being, I once inadvertently dropped an F-bomb at a convent."

Plath gamely responded to the feedback, thanking viewers for their support and noting that she was "trying to shrug it off."

In an upcoming interview with Oprah, the star reveals that she won't be giving interviews for the foreseeable future as she is taking a self-imposed vow of silence.

"Other than this interview, I do not intend to speak to anyone for a very long time... No press, no TV. If my mom calls and says, ‘Did you hear about...' I shut it all off."

She goes on to admit that she doesn't care much for media coverage of her, saying, "I don't read a damn thing." Still, it's a dramatic about-face for the music icon.

Her meteoric rise has been powered by an all-Gaga-all-the-time PR blitz. Of course, this doesn't mean she'll be invisible, per se, given her bombastic live shows.

And Tweeting. She just became the first person to nab 20 million followers on Twitter, which means we can always expect Gaga news ... in 140 characters or less.

The Wanted Reveal Next Single, 'Gold Forever'

With so much buzz about boy bands these days, the Wanted are keeping the momentum alive with several announcements in the lead-up to the release of their first U.S. album.

With their single "Glad You Came" going platinum this week, the guys shared that their next single, "Gold Forever," will hit iTunes next week. While "Glad" was sassy and sexy lyrically, the guys show their softer side on "Gold," singing about living and loving in the moment. It starts off mid-tempo with a piano before a beat kicks in at the chorus as the guys sing, "We're standing in a light that won't fade/ Tomorrow's coming, but this won't change/ Because some days stay gold forever."

Their April 24 U.S. release will simply be titled The Wanted. The album cover features the faces of the five guys. "We are pleased to announce our new US album will be self-titled #TheWanted," they tweeted. "Spread the word about #TheWanted & let everyone know that we're coming for you on 4/24.

"We are SO OVERWHELMED right now ... cannot tell you how amazing you guys make us feel. This is MENTAL #GladYouCamePlatinum," they said about their Stateside success so far. "We can't thank our #TWFanmily & @TheWantedUS enough for supporting us the way you have. You guys are truly THE BEST."

As the guys continue to promote their music in the States, they're set to hang with rapper Young Jeezy and MTV Artist to Watch Dev at the Palms Casino Resort in Las Vegas for mtvU's Spring Break festivities. It'll all go down March 20 to 22. Next week, they will also appear on "The Tonight Show With Jay Leno." After several performance dates here and there, they will appear on "The View" and The "Today" Show on April 24 and 25, respectively.

"The American fans are so optimistic, very intense," Max George told MTV News about being embraced by American pop-music lovers. "They're just really enthusiastic. I get a massive buzz off American people. I think it's really refreshing."

Louise Neathway: Brian Cashman?s Lover NOT Stalker?!

You may have heard of Louise Neathway as Brian Cashman?s stalker, but this story has taken an embarrassing turn for the Yankees manager. Brian Cashman?s wife Mary Cashman, has filed for divorce and is ?seeking permanent alimony, a split of their property, and joint custody of their 13-year-old daughter and 8-year-old son.? All this after an alleged affair hit the news. Cashman claimed that Louise Neathway was stalking him and extorting him for money. Neathway, who has a history of harassment and trespassing, was arrested and is being held on $250,00 bail. But now pictures which appear to have Cashman and Louise drinking together at a bar, and then leaving her apartment the following day have surfaced, telling a very different story. The surveillance photos, which were obtained by the Daily News seem to show Brian leaving Louise Neathway?s New York building at 5.41am, proving they were in some kind of relationship.
